KE00972 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human GATA3 ELISA kit is to be used to detect and quantify protein levels of endogenous human GATA3. The assay recognizes human GATA3. An antibody specific for human GATA3 has been pre-coated onto the microwells. The human GATA3 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human GATA3 is added to detect the captured human GATA3 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
GATA3 is a member of a family of transcriptional regulators with the capacity to function in determination of cell identity. It is known for its ability to bind to the DNA sequence "GATA", which enables its key role in critical processes in multiple tissues and organs, including the immune system, endocrine system, and nervous system. GATA3 also modulates cell differentiation, proliferation, and apoptosis by controlling gene expression. Its expression is associated with cell type specification in the immune system, where its action is integral to the differentiation of multiple cell types. Dysregulation of GATA3 is implicated in several diseases, such as breast cancer, hematological malignancies like T-cell lymphomas, autoimmune diseases, and neurodegenerative disorders.
