Product Information

KE00600 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human FAP ELISA kit is to be used to detect and quantify protein levels of endogenous human FAP. The assay recognizes human FAP. An antibody specific for human FAP has been pre-coated onto the microwells. The human FAP protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human FAP is added to detect the captured human FAP protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

FAP, also known as fibroblast activation protein-alpha (FAP-alpha) is a cell surface glycoprotein serine protease, which can participate in extracellular matrix degradation and is involved in many cellular processes including tissue remodeling, fibrosis, wound healing, inflammation and tumor growth. FAP is a single transmembrane protein that locates the cytoplasm and plasma membrane. Immunohistochemical studies have shown that FAP is transiently expressed in certain normal fetal mesenchymal tissues but that normal adult tissues as well as malignant epithelial, neural, and hematopoietic cells are generally FAP-negative. However, most of the common types of epithelial cancers contain abundant FAP-reactive stromal fibroblasts.
