Product Information
KE00929 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human ETS2 ELISA kit is to be used to detect and quantify protein levels of endogenous human ETS2. The assay recognizes human ETS2. An antibody specific for human ETS2 has been pre-coated onto the microwells. The human ETS2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human ETS2 is added to detect the captured human ETS2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
ETS2 (Protein C-ets-2), a member of the ETS family of transcription factors, plays a key role in transcriptional regulation and cellular functions such as proliferation, differentiation, migration, transformation, and apoptosis. ETS2 binds to the GGA(A/T) motif (Ets-binding site) in gene promoters to regulate target gene expression. In placental development, ETS2 is essential for controlling trophoblast differentiation via transcriptional regulation of genes like human chorionic gonadotropin β5 (hCGβ5), with its activity modulated by the Ras/MAPK pathway. In immune regulation, ETS2 suppresses inflammatory cytokine production (e.g., IL-6) in macrophages via MAPK/NF-κB signaling and directly binds to the IL-6 promoter. Additionally, ETS2 is critical for lipopolysaccharide (LPS)-induced miR-155 expression (a microRNA involved in inflammation and oncogenesis), and this induction is inhibited by interleukin-10 (IL-10) through suppression of ETS2.
