KE00731 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human DR5 ELISA kit is to be used to detect and quantify protein levels of endogenous human DR5. The assay recognizes human DR5. An antibody specific for human DR5 has been pre-coated onto the microwells. The human DR5 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human DR5 is added to detect the captured human DR5 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Death receptor 5 (DR5) is a member of the tumor necrosis factor receptor superfamily. It plays a critical role in regulating apoptosis through its interaction with the TNF-related apoptosis-inducing ligand (TRAIL). When DR5 is activated by TRAIL, it triggers a cascade of intracellular signaling events that lead to the activation of caspases, the proteases responsible for executing apoptosis. This selective targeting of cancer cells by DR5 has made it a promising candidate for cancer therapy. Elevated levels of soluble DR5 have been observed in the serum of cancer patients, indicating its potential as a biomarker.
