KE00124 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Cystatin C ELISA kit is to be used to detect and quantify protein levels of endogenous human . The assay recognizes human Cystatin C. An antibody specific for human Cystatin C has been pre-coated onto the microwells. The human Cystatin C protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for human Cystatin C is added to detect the captured human Cystatin C protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Cystatin C is a 13-kDa inhibitor of cysteine proteinases which is secreted by all cell types and is completely cleared from the organism through glomerular filtration, shown to be an early and sensitive biomarker of renal dysfunction. It is also used as an emerging biomarker in cardiovascular disease. Cystatin C is involved in a variety of inflammatory reactions. The concentration of serum cystatin C has also been shown to be unaltered in certain inflammatory conditions or other disorders of metabolism. The plasma level of serum cystatin C can be expressed as its level of generation from cells and diet and its subsequent elimination through the gut, liver, and kidneys.
