Product Information
KE00865 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Caspase 10 ELISA kit is to be used to detect and quantify protein levels of endogenous human Caspase 10. The assay recognizes human Caspase 10. An antibody specific for human Caspase 10 has been pre-coated onto the microwells. The human Caspase 10 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Caspase 10 is added to detect the captured human Caspase 10 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Caspase-10 is a protein closely related to apoptosis. It is highly homologous to Caspase-8. As the initiator caspase, Caspase-10 plays an important role in the apoptosis pathway mediated by death receptors (such as Fas, TNF-α receptor, etc.). It is recruited and autocatalytically activated in the death-inducing signaling complex (DISC), and then activates downstream effector caspases (such as Caspase-3, -6 and -7), thereby initiating apoptosis. CASP10 deficiency is the cause of autoimmune lymphoproliferative syndrome type 2A (ALPS2A), familial non-Hodgkin's lymphoma (NHL) and gastric cancer (GASC). The full length consists of a 219-amino acid propeptide and 6 isoforms produced by alternative splicing.
