Product Information
KE00451 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human CDK2 ELISA kit is to be used to detect and quantify protein levels of endogenous human CDK2. The assay recognizes human CDK2. An antibody specific for human CDK2 has been pre-coated onto the microwells. The human CDK2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human CDK2 is added to detect the captured human CDK2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Cyclin-dependent kinase 2 (CDK2) is also named as CDKN2. CDK2 is a core cell-cycle kinase that phosphorylates many substrates to drive progression through the cell cycle. CDK2 is hyperactivated in multiple cancers and is therefore an attractive therapeutic target. CDK4/6-cyclin D complexes phosphorylate and inactivate the retinoblastoma protein (Rb1), liberating E2F to promote transcription of cyclins E and A, which activate CDK2. Activated CDK2 hyperphosphorylates Rb1, establishing a potent positive feedback loop that ensures the continued expression of cyclins E and A, as well as other proteins that are essential for S phase.
