KE00505 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human BCL9 ELISA kit is to be used to detect and quantify protein levels of endogenous human BCL9. The assay recognizes human BCL9. An antibody specific for human BCL9 has been pre-coated onto the microwells. The human BCL9 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human BCL9 is added to detect the captured human BCL9 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
BCL9 is a key developmental regulator and a well-established oncogenic driver in multiple cancer types. BCL9 is expressed in a variety of tissues, including the thymus, spleen, heart, skeletal muscle, and lungs. BCL9 promotes tumor progression and metastasis by enhancing the Wnt/β-catenin signaling pathway. It also plays a role in the regulation of immune cell infiltration in tumors, contributing to immune evasion.
