Product Information
KE00774 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human BCL3 ELISA kit is to be used to detect and quantify protein levels of endogenous human BCL3. The assay recognizes human BCL3. An antibody specific for human BCL3 has been pre-coated onto the microwells. The human BCL3 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human BCL3 is added to detect the captured human BCL3 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
BCL3 (B-cell lymphoma 3 protein) has seven ankyrin repeat domains, a proline-rich N terminal domain and a serine- and proline-rich C-terminal domain. BCL3 is an important contributor to chronic inflammatory disease states, such as osteoarthritis and rheumatoid arthritis. BCL3 is a predominantly nuclear protein containing a trans-activation domain, and it can be recruited to NF-κB-responsive promoters, resulting in transcriptional activation or repression depending on the subunit composition of NF-κB complexes. BCL3 is known to be overexpressed in human breast cancers and is associated with reduced metastasis-free survival.
