Product Information
KE00855 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Alpha-2-macroglobulin ELISA kit is to be used to detect and quantify protein levels of endogenous human Alpha-2-macroglobulin. The assay recognizes human Alpha-2-macroglobulin. An antibody specific for human Alpha-2-macroglobulin has been pre-coated onto the microwells. The human Alpha-2-macroglobulin protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Alpha-2-macroglobulin is added to detect the captured human Alpha-2-macroglobulin protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Alpha-2-Macroglobulin, also known as α2-macroglobulin (α2M or A2M), is a protein abundant in the plasma of vertebrates and several invertebrates. A2M is an evolutionarily conserved arm of the innate immune system. It also mediates the proliferation of T cells and macrophages. A2M acts as a nonspecific protease inhibitor involved in the host defense mechanism that inactivates both endogenous and exogenous proteases, including trypsin, thrombin and collagenase. Even though A2M is produced predominantly by the liver, it may also be expressed in the reproductive tract, heart, and brain, and may have important roles in many physiological processes and medical illnesses including Alzheimer's disease.
