Product Information
KE00332 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human AGT ELISA kit is to be used to detect and quantify protein levels of endogenous human AGT. The assay recognizes human AGT. An antibody specific for human AGT has been pre-coated onto the microwells. The human AGT protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human AGT is added to detect the captured human AGT protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Angiotensinogen(AGT), a precursor of angiotensin II (Ang II), is expressed and synthesized largely in the liver and cleaved by the enzyme renin in response to lowered blood pressure. It has a key role in mediating vascular constriction and regulating salt and fluid homeostasis. The resulting product, angiotensin I, is then cleaved by angiotensin converting enzyme (ACE) to generate the physiologically active enzyme angiotensin II. Mutations in this gene are associated with susceptibility to essential hypertension, and can cause renal tubular dysgenesis, a severe disorder of renal tubular development.
