Product Information
KE00660 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human AES ELISA kit is to be used to detect and quantify protein levels of endogenous human AES. The assay recognizes human AES. An antibody specific for human AES has been pre-coated onto the microwells. The human AES protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human AES is added to detect the captured human AES protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Amino-terminal enhancer of split (AES), also known as TLE5 or Grg5, belongs to the groucho/TLE family of proteins. AES can function as a homooligomer or as a heteroologimer with other family members in order to forcefully repress the expression of other family member genes. AES can repress NFkB-regulated gene expression and is believed to have a significant role in initiating and maintaining cell differentiation events. AES has been found to suppress local invasion and intravasation through inhibition of the Notch pathway, leading to preventing the metastatic spread of endogenous tumors, such as colon cancer and prostate cancer.
