Product Information
KE00809 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human 4EBP1 ELISA kit is to be used to detect and quantify protein levels of endogenous human 4EBP1. The assay recognizes human 4EBP1. An antibody specific for human 4EBP1 has been pre-coated onto the microwells. The human 4EBP1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human 4EBP1 is added to detect the captured human 4EBP1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
4EBP1 is a translational regulator and functions by sequestering eukaryotic translation initiation factor 4E (eIF4E) from the translation initiation machinery. 4E-BP1 contains at least six phosphorylation sites (Thr37, Thr46, Ser65, Thr70, Ser83, and Ser112), four of which (Thr37, Thr46, Ser65, and Thr70) are known to be regulated by mTOR signaling. Phosphorylation of Thr37 and Thr46 is thought to prime 4E-BP1 for sequential phosphorylation of Ser65 and Thr70, which results in the dissociation of 4E-BP1 from eIF4E. (PMID: 12747827)
