Background Information
HELIOS, encoded by the IKZF2 gene (Ikaros Family Zinc Finger Protein 2), is a transcription factor belonging to the Ikaros family of zinc-finger proteins. It is primarily expressed in hematopoietic cells, particularly in regulatory T cells (Tregs) and early lymphoid progenitors, where it plays a critical role in immune regulation, T-cell development, and the maintenance of Treg suppressive function. HELIOS binds to specific DNA sequences to regulate gene expression involved in lymphocyte differentiation and immune homeostasis. It is also widely used as a stable marker for activated Tregs. Dysregulation of HELIOS has been implicated in autoimmune diseases, allergy, and cancer immunotherapy resistance.
