Background Information
Guanine nucleotide exchange factor H1 (GEF-H1) is also named ARHGEF2 and LFP40. GEF-H1, encoded by ARHGEF2 gene, is a member of the larger group of DH/PH containing GEFs (PMID: 15688002). GEFH1 is a RhoA activator that is associated with microtubules (MT) and its localization and activity are regulated, in part, by MT and fibronectin-binding integrins (PMID: 40154811). GEF-H1 is a RhoA specific MT-associated GEF, and can be activated by MT-depolymerizing agents such as nocodazole and colchicine. In epithelial cells, GEF-H1 is necessary for RhoA-dependent contractility and stress fiber formation in response to MT depolymerization (PMID: 29188751). In polarized epithelial cells, GEF-H1 is sequestered to apical junctions (AJ) by binding to the AJ structural proteins cingulin or paracingulin (PMID: 33126816).
