Background Information
The FAM21A protein, full name Family with sequence similarity 21 member A, belongs to the FAM21 protein family. This protein is a core subunit of the WASH complex (Wiskott-Aldrich syndrome protein and SCAR homolog complex) and plays a critical role in endosomal sorting and membrane trafficking. Its primary function is to recruit the WASH complex to the endosomal membrane surface by binding to phospholipids (such as phosphatidylinositol 4-phosphate) and the VPS35 subunit of the Retromer complex, thereby anchoring the complex to the correct position on the endosomal membrane.
