Background Information

Cerebral dopamine neurotrophic factor (CDNF) is a secreted protein belonging to the mesencephalic astrocyte-derived neurotrophic factor (MANF) family. Unlike classical neurotrophic factors, CDNF is primarily localized in the endoplasmic reticulum and exerts neuroprotective effects through modulating the unfolded protein response (UPR). CDNF has demonstrated potent protective effects on dopaminergic neurons in preclinical models of Parkinson's disease (PD), reducing neuroinflammation and alpha-synuclein aggregation. Its unique mechanism-targeting ER stress rather than receptor tyrosine kinases-offers a promising therapeutic avenue for neurodegenerative disorders. Clinical trials are currently evaluating recombinant human CDNF as a disease-modifying therapy for PD (PMID: 34284712; 37555005; 38290651).
