Background Information
ABCB8 is a mitochondrial inner membrane protein and a member of subfamily B of the ABC transporter superfamily. It contains two transmembrane domains (TMDs) and two nucleotide-binding domains (NBDs) and transports substrates in an ATP-dependent manner. This protein is a core regulator of mitochondrial iron homeostasis and is responsible for transporting iron-sulfur cluster (Fe-S) precursors from the mitochondrial matrix to the cytosol, thereby maintaining cellular iron homeostasis. Gene knockout or loss of function leads to mitochondrial iron accumulation, reactive oxygen species (ROS) burst, respiratory chain dysfunction, and apoptosis. In addition, ABCB8 is closely associated with iron metabolism-related diseases such as cardiomyopathy and Friedreich's ataxia, and is an important molecule in studies of mitochondrial iron transport, oxidative stress, and tumor metabolism.
