Background
ROBO1 (Roundabout Guidance Receptor 1) is a type I single-pass transmembrane glycoprotein of the immunoglobulin superfamily, serving as the receptor for SLIT1/2 ligands. It mediates axon guidance, neuronal migration, and angiogenesis during development. Dysregulated ROBO1 drives tumor invasion/metastasis in multiple cancers and pathological ocular neovascularization. It is a preclinical therapeutic target; neutralizing antibodies and small molecules blocking SLIT-ROBO1 signaling are under investigation for cancer and eye diseases.
