Product Information

KE00084 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The PGA5 ELISA kit is to be used to detect and quantify protein levels of endogenous PGA5. The assay recognizes human PGA5. An antibody specific for PGA5 has been pre-coated onto the microwells. The PGA5 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody specific for PGA5 is added to detect the captured PGA5 protein. For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Pepsinogen I belongs to the peptidase A1 family. The gene endoces a 388 amino acid protein with a 15 amino acid signal peptide and 46 amino acid propeptide. Pepsinogen I is secreted only by the chief cells of the corpus, and its serum level decreases with increasing grade of the corpus atrophy (loss of oxyntic glands). Low serum levels of pepsinogen I are indicative of chronic atrophy, a risk factor for gastric cancer.
