Product Information

KE00117 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The P-cadherin ELISA kit is to be used to detect and quantify protein levels of endogenous P-cadherin. The assay recognizes human P-cadherin. An antibody specific for P-cadherin has been pre-coated onto the microwells. The P-cadherin protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for P-cadherin is added to detect the captured P-cadherin protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Cadherins are a family of transmembrane glycoproteins that mediate calcium-dependent cell-cell adhesion and play an important role in the maintenance of normal tissue architecture. P-cadherin (placental cadherin), also known as CDH3 (cadherin 3), is a classical member of the cadherin superfamily which also include E-, N-, R-, and B-cadherins. Aberrant expression of P-cadherin has been reported in many cancers, including breast, gastric, endometrial, ovarian, and prostate cancer. P-cadherin can be cleaved into a soluble fragment (soluble P-cadherin), which has been found in human milk, semen, serum, and nipple aspirate fluid (NAF).
