KE00284 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human CD16 ELISA kit is to be used to detect and quantify protein levels of endogenous CD16. The assay recognizes human CD16. An antibody specific for CD16 has been pre-coated onto the microwells. The CD16 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for CD16 is added to detect the captured CD16 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

CD16, also known as the low affinity Fc gamma receptor III for IgG (FcγRIII), exists in two isoforms, FcγRIIIa (CD16a) and FcγRIIIb (CD16b), encoded by two nearly identical genes, FCGR3A and the FCGR3B. CD16a is expressed on NK cells, macrophages, and placental trophoblasts as a polypeptide-anchored transmembrane protein while CD16b is expressed on neutrophils in a glycosylphosphatidylinositol (GPI)-anchored form. CD16a forms a heteromeric structure with the Fc epsilon RI (gamma) and/or CD3 (zeta) subunits. CD16 mediates antibody-dependent cellular cytotoxicity (ADCC) and other antibody-dependent responses, such as phagocytosis. CD16 can be cleaved and released as a soluble form (sCD16).
