Background Information

F-box proteins have been shown to be critical for the ubiquitin-mediated degradation of cellular regulatory proteins, and they are a family of eukaryotic proteins characterized by an approximately 40 amino acid motif. SCF complex, a class of ubiquitin ligases, consists of invariable components, Skp1 and Cullin, and variable components of F-box proteins, which have a primary role in determining substrate specificity. FBXW5 (F-box/WD repeat-containing protein 5) is a novel protein containing WD-40 repeats, which were frequently involved in protein-protein interactions. FBXW5 negatively regulates TAK1 in the IL-1beta signaling pathway. By forming different E3 ubiquitin-protein ligase complexes, FBXW5 mediates the polyubiquitination and subsequent degradation of TSC2 and centriolar protein HsSAS-6.
