Product Information

KE00069 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The CYB5R3 ELISA kit is to be used to detect and quantify protein levels of endogenous CYB5R3. The assay recognizes human CYB5R3. An antibody specific for CYB5R3 has been pre-coated onto the microwells. The CYB5R3 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody specific for CYB5R3 is added to detect the captured CYB5R3 protein. For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

CYB5R3, also named as DIA1 and B5R, belongs to the flavoprotein pyridine nucleotide cytochrome reductase family and has two naturally occuring isoforms. Isoform 1 is anchored to the cytoplasmic side of the endoplasmic reticulum membrane and mitochondrion outer membrane, while isoform 2 is the soluble form found in red blood cells. There are two types of methemoglobinemia CYB5R3-related. In type 1, the defect affects the soluble form of the enzyme, is restricted to red blood cells, and causes well-tolerated methemoglobinemia. In type 2, the defect affects both the soluble and microsomal forms of the enzyme and is thus generalized, affecting red cells, leukocytes and all body tissues. Type 2 methemoglobinemia is associated with mental deficiency and other neurologic symptoms. The ELISA kit is suitable for testing serum, plasma, and cell lysates.
